History
DMEM (Dulbecco’s Modified Eagle Medium) and F12 (Ham’s F-12 Nutrient Mixture) are two widely used basal media for cultivating mammalian cells in tissue culture. Both were developed to support the growth and maintenance of a wide range of cell types under in vitro conditions.
History: DMEM was initially formulated by Dulbecco and Vogt in 1954 for the polio virus studies in embryonic mouse cells. Over the years, it has been modified and enriched to cater to a diverse array of mammalian cells. F12, on the other hand, was developed by Ham in the 1960s for the serum-free growth of cells derived from the Chinese hamster ovary.
Recognizing that certain cells require components from both DMEM and F12 for optimal growth, researchers started mixing these two media, leading to the creation of the DMEM/F12 blend. This combination harnesses the unique qualities of both media, offering a broader range of nutrients, thus expanding the spectrum of cells that can be supported.
Usage: DMEM/F12 finds its application predominantly in the culture of stem cells, primary cells, and some specific cell lines. It’s especially favored when cells necessitate a medium that provides a delicate balance between the high nutrient levels in DMEM and the specialized components in F12.
A notable example of its application is in the cultivation of neural stem cells and brain organoids, where DMEM/F12 provides an environment that closely mimics the in vivo conditions of the brain. Moreover, with the addition of specific growth factors and supplements, DMEM/F12 can be tailored to support the differentiation of pluripotent stem cells into various lineages.
In summary, DMEM/F12 plays a pivotal role in tissue culture by offering a versatile and nutrient-rich medium, bridging the gap between DMEM and F12, and facilitating the growth and differentiation of a myriad of cell types.
